Abstract

Leptin isapolypeptidesecretedbyadipocyteswhichbindstoaspecificreceptor(Ob-R)thatisexpressed in varioustissues.ThewidedistributionoftheOb-Rsuggeststhatleptinmightexertdiversebiological functions, notonlybyregulatingenergymetabolismandappetite,butalsobyactingasamitogenin many celltypes,includingkeratinocytes. In thisstudy,thepresenceandlocalizationofOb-RwasinvestigatedintheskinofthedogusingRT-PCR and immunohistochemicaltechniques.RT-PCRrevealedthepresenceofOb-Rm-RNAintheskinspeci- mens collectedfromthedorsalregionoftwosmoothcoatbreeddogs.Throughimmunohistochemistry performed ontheskinoffivedogs,theexpressionofthereceptorwasobservedinthebasallayerofthe epidermis, inthehairfolliclesaswellasintheapocrinesweatandsebaceousglands.NostainingforOb-R was detectedinthesuprabasalepidermislayers.Strongpositivesignalswereobservedinmanycellsof the outerrootsheathofhairfolliclesingrowingandinregressivephases.TheidentificationofOb-Rin the abovetargetssuggeststhatleptinmayplayaroleintheregulationofcyclicrenewaloftheepidermis and skinappendagesindog.Thisstudyrepresentsanimportantcontributiontounderstandthecomplex mechanisms thatareinvolvedintheskinbiologyinthisspecies.
