Mucins are critical cytoprotective glycoproteins and alterations of epithelial gastric mucins have been described in different pathological conditions. We evaluated histo- ‘and immunohistochemically mucins modifications in two major types of canine gastric cancer, and putative usefulness of mucins in understanding the progression of these gastriC neoplasia.), Since 2009, 16 formalin-fixed paraffin-embedded gastric cancer biopsy specimens and surgically resected portions of dog stomach were sectioned at our laboratory. According to WHO, samples were classified as Signet Ring Cell Carcinoma type (SRCC; 8 samples), and Tubular Adenocarcinoma (TAC; 8 samples). Alcian Blue (AB) staining was performed in order to identify sialomucins (staining blue at pH 2.5) and sulfomucins (staining brown at pH 1.0) while Periodic acid-Schiff’s (PAS) staining was performed for neutral mucins (staining magenta). In general, a significant increase of acidic mucins was found in gastric carcinoma. MUC1, MUC2, Ga11, and Gal3 antibodies were used (by himmunohistochemistry) for gastric mucins characterization. In normal gastric mucosae we found that neutral mucins were predominant, whereas sulfomucins and sialomucins were found to be very highly expressed in TAC. Similarly, SRCC showed a significant staining of sialomucins rather than sulfomucins, with a moderate staining of neutral mucins in intracytoplasmic vacuoles., Levels of gastric mucin antigen MUC1, was decreased in both SRCC and TAC , as how de novo expression of the intestinal mucin MUC2 in goblet cells was decresed in T AC forms. Gal-1 was constantly unexpressed in both forms, and Gal-3 was detected in cytoplasm and nuclei of the gastric cancer celis, independently to the morphological type of the tumor. We suggest that sulfomucins have a greater role in gastric carcinogenesis than the neutral mucins, moreover we support that sialomucins and sulfomucins play an important role in cancer progression and metastasis.
