The aim of the present study was to evaluate in the horse Leydig cells 1) the presence of kisspeptin (KiSS) and gonadotropin-releasing hormone (GnRH) and their cognate receptors (GPR54 and GnRHR, respectively) and 2) the possible direct interaction between KiSS and GnRH on the testicular endocrine activity. KiSS, GPR54, GnRH and GnRHR were immunolocalized in the cytoplasm of Leydig cells. KiSS and GnRH increased testosterone and PGF2 and decreased PGE2 in vitro secretions of Leydig cells. GnRH antagonist counteracted the KiSS effects, whereas KiSS antagonist did not affect the GnRH effects. The present study indicates that KiSS/GPR54 and GnRH/GnRHR systems are present in horse Leydig cells and that both peptides affect the testicular endocrine activity. In particular, the KiSS effects are mediated by GnRH, so suggesting that the hypothalamic interaction between KiSS and GnRH occurs also in horse testis.
