Objectives: The aim of the study was to evaluate the mechanism beyond the dopaminergic alteration in striatum observed after early life exposure to permethrin insecticide. Methods: Pup rats have been exposed to oral permethrin treatment (34 mg/kg) from 6th to 21st of life. Biochemical analysis of dopaminergic level and gene expression and DNA methylation of Nurr1 in striatum of adolescent rats were performed. Results: Dopamine level appears decreased in striatum from rats treated during early life with permethrin. The decrease of Nurr1 gene expression in striatum of adolescent rats treated with permethrin could play an important role in the dopamine depletion. Increased promoter methylation of Nurr1 is the cause of dopaminergic alteration observed in treated rats. These changes also influenced negatively the cognitive behaviour of animals.
